I caught a queen Camoonotus Pennsylvanicus on 5/25/2024 and am curious if it's fertile. It still keeps its wings, but that does not indicate whether it's haploid. It also has been ripping the cotton from its test tube to escape(unsuccessful), does this mean its not rearing brood?

If you just caught her today, that's pretty typical. Give her like a week in the dark and then check on her. After a few days you should see eggs if she's fertile, but I usually give them a week or two to settle and not stress them.
